The antibody-conjugated bead set is designed to be used in conjunction with 's Histone H3 PTM Multiplex Kit . Follow the recommendations in the Histone H3 PTM Multiplex Kit manual for the preparation and use of the antibody-conjugated bead set. Each analyte specific antibody-conjugated bead contains its own unique fluorescent label allowing the user to distinguish different bead sets within a single sample for multiplex experiments. The beads can be read and identified using a Luminex 200?, MAGPIX? or FLEXMAP 3D? instrument and xPONENT? software. The associated bead region for the Histone H3K56ac Ab-conjugated bead set is #75. The recommended detection limit for this bead set is: Acid extracts - use greater than 125 ng per well of sample Purified histones - use greater than 7.8 ng per well of sample Recommendations for Dilution of the Biotinylated Histone H3 Antibody: H3K56ac Ab-conjugated Beads may benefit from the use of Biotinylated Histone H3 antibody at higher concentrations in order to increase the reporter signal. If the H3K56ac Ab-conjugated Bead generates MFI values less than 1000 in 1 μg lysate, it is recommended to use the Biotinylated Histone H3 reporter antibody at 1:250 dilution.
